General description
α-Ketoglutarate Dehydrogenase (α-KGDH, EC 1.2.4.2) is a highly regulated Krebs cycle enzyme, forms the E1 subunit of the mitochondrial 2-oxoglutarate dehydrogenase complex. It catalyzes the conversion of α-ketoglutarate to succinyl-CoA and produces NADH directly providing electrons for the respiratory chain. Decreased α-KGDH complex activity has been associated with oxidative stress and neurodegenerative disorders such as Alzheimer′s and Parkinson′s diseases.
Principle
The α-Ketoglutarate Dehydrogenase Activity Colorimetric Assay Kit provides a simple procedure for measuring α-ketoglutarate dehydrogenase activity in a variety of tissues, cells, and isolated mitochondria. α-KGDH activity is determined by measuring a colorimetric product with absorbance at 450 nm (A450) proportional to the enzymatic activity present. One unit of α-ketoglutarate dehydrogenase is the amount of enzyme that generates 1.0 µmole of NADH per minute at pH 7.5 at 37 °C.
